Somatic Cell Mutation Assay at the Glycophorin-a Locus in Human Erythrocytes for Biodosimetry

چکیده

For risk assessment, it is necessary to evaluate the dose of human exposure to radiation. Ionizing radiation being an environmental genotoxic factor induces dosedependent somatic mutations. Recent studies support the somatic-mutation theory of carcinogenesis. Mutations at the Glycophorin–A (GPA) loci have been studied for establishment of somatic mutation assays. GPA assay provides a lifetime biological dosimetry technique to detect exposure to radiation as the mutations are accumulated in the long lived hematopoietic stem cells. In this review, we present the various concepts and advancements in the GPA mutational assay, and its role in biodosimetry and risk assessment.

Detection of somatic mutations at the glycophorin A locus in erythrocytes of atomic bomb survivors using a single beam flow sorter.

A modified method was developed for measuring the frequency of variant erythrocytes at the glycophorin A locus using a single beam cell sorter (SBS). Fluorescein- or phycoerythrin-labeled monoclonal antibodies specific for the M or N glycophorin A alleles were used for the SBS assay. To prevent contamination of nucleated cells in the sorting windows, the nucleated cells in the fixed erythrocyte...

Human somatic mutation assays as biomarkers of carcinogenesis.

This paper describes four assays that detect somatic gene mutations in humans: the hypoxanthine-guanine phosphoribosyl transferase assay, the glycophorin A assay, the HLA-A assay, and the sickle cell hemoglobin assay. Somatic gene mutation can be considered a biomarker of carcinogenesis, and assays for somatic mutation may assist epidemiologists in studies that attempt to identify factors assoc...

Glycophorin A Gene Mutations of Atomic Bomb Survivors Cancer Susceptibility: Prospective Study on Erythrocyte Individual Variation of Somatic Gene Mutability in Relation to

It has previously been reported that hemizygous mutant fraction (Mf) at the glycophorin A (GPA) locus in erythrocytes increased with radiation dose in heterozygotes among Hiroshima and Nagasaki atomic bomb survivors. In the present study, we analyzed the relationship between GPA Mf and cancer risk using newly developed cancers among previously cancer-free subjects whose GPA Mf had been measured...

Cytokinesis blocked micronucleus (CB-MN) assay for biodosimetry of high dose accidental exposure

Background: Most of the cytogenetic dosimetry, except Premature Chromosome Condensation assay, saturates at 6 Gy. Present studies are aimed to assess feasibility of simple CB-MN assay for prescreening of high dose accidental exposures. Materials and Methods: Human peripheral blood lymphocytes from two donors were gamma irradiated to different doses ranging from 0-15Gy, cultured for 72 h. Cytoch...
